A highly selective and efficient LC–MS/MS method was developed to determine the plasma concentration of magnolol, hesperidin, neohesperidin and geniposide following oral administration of Zhi‐Zi‐Hou‐Po decoction in normal and depressed rats. Plasma samples were pretreated by protein precipitation with methanol. Chromatographic separation was performed on an XTerra® MS C18 column using a gradient elution with a mobile phase composed of acetonitrile–0.1% aqueous formic acid. The proposed method was validated to be specific, accurate and precise for the analytes determination in plasma samples. The calibration curves displayed good linearity over definite concentration ranges for the analytes. The intra‐ and inter‐day precision of the proposed method at three different levels were all within <11.13% and the relative errors ranged from ?8.46 to 8.93%. The recovery of the four compounds ranged from 82.72 to 89.08% and no apparent matrix effect was observed during sample analysis. After full validation, the established method was successfully applied for comparing the pharmacokinetics of four components between normal and depressed rats. The results showed that the AUC and Cmax of four analytes in depressed rats were significantly different from those in normal rats and might provide helpful information to guide the clinical use of Zhi‐Zi‐Hou‐Po to treat depression.  相似文献

The development of highly sensitive and selective methods for the detection of lead ion (Pb2+) is of great scientific importance. In this work, we develop a new surface‐enhanced Raman scattering (SERS)‐based sensor for the selective trace measurement of Pb2+. The SERS‐based sensor is assembled from gold nanoparticles (AuNPs) and graphene using cucurbit[7]uril (CB[7]) as a precise molecular glue and a local SERS reporter. Upon the addition of Pb2+, CB[7] forms stronger complexes with Pb2+ and desorbs from AuNPs, resulting in a sensitive “turn‐off” of SERS signals. This SERS‐based assay shows a limit of detection (LOD) of 0.3 nm and a linear detection range from 1 nm to 0.3 μm for Pb2+. The feasibility of the assay is further demonstrated by probing Pb2+ in real water samples. This SERS‐based analytical method is highly sensitive and selective, and therefore holds promising applications in environmental analysis.  相似文献

Without excess Li, anode-free Li-metal batteries (AFLMBs) have been proposed as the most likely solution to realizing highly-safe and cost-effective Li-metal batteries. Nevertheless, short cyclic life puzzles conventional AFLMBs due to anodic dead Li accumulation with a local current concentration induced by irreversible electrolyte depletion, insufficient active Li reservoir and slow Li+ transfer at the solid electrolyte interphase (SEI). Herein, SrI2 is introduced into carbon paper (CP) current collector to effectively suppress dead Li through synergistic mechanisms including reversible I/I3 redox reaction to reactivate dead Li, dielectric SEI surface with SrF2 and LiF to prevent electrolyte decomposition and highly ionic conductive (3.488 mS cm−1) inner layer of SEI with abundant LiI to enable efficient Li+ transfer inside. With the SrI2-modified current collector, the NCM532/CP cell delivers unprecedented cyclic performances with a capacity of 129.2 mAh g−1 after 200 cycles.  相似文献

2-(3'-羟基-4'-甲氧基苯基)-3-(3",4",5"-三甲氧基苯甲酰基)-6-甲氧基吲哚(OXi8006)能够有效抑制微管蛋白聚合,而表现出良好抗癌活性.目前报道的OXi8006全合成路线较长、总收率低,且反应条件苛刻.为了更高效地合成该化合物,从而为进一步的活性和构效关系研究提供原料.以廉价易得的异香兰素为起始原料,先合成芳基乙炔,再与3,4,5-三甲氧基苯甲醛通过亲核加成、氧化反应获得二芳基炔酮、二芳基炔酮再与邻碘代苯胺通过杂迈克尔加成和分子内Heck反应构建出OXi8006的主体结构——2-芳基-3-芳酰基取代吲哚,从而缩短了合成路线,并使总收率提高到20%.  相似文献

Photoreactivation is an error‐free mechanism of DNA repair, utilized by prokaryotes and most eukaryotes and is catalyzed by specific enzymes called DNA photolyases. Photoreactivation has been reported in Vibrio parahaemolyticus WP28; however, information on photolyases in V. parahaemolyticus (V.p) strains has not been reported. This study examined the photoreactivation in V.p RIMD2210633. The photolyase responsible for repairing cyclobutane pyrimidine dimer (CPD) in DNA was identified, and the corresponding gene was determined as VPA1471. The protein was overexpressed in Escherichia coli and was purified for functional assessment in vitro. The mRNA level and protein expression level of this gene increased after ultraviolet A (UVA) illumination following ultraviolet C (UVC) irradiation. In vitro experiments confirmed that the protein encoded by VPA1471 could reduce the quantity of CPD in DNA. We designated the corresponding gene and protein of VPA1471 phr and Phr, respectively, although the function of two other photolyase/cryptochrome family members, VPA0203 and VPA0204, remains unclear. UV (ultraviolet) irradiation experiments suggest that these two genes possess some photorepairing ability. Therefore, we hypothesize that VPA0203 and VPA0204 encode (6‐4) photolyase in V. parahaemolyticus RIMD2210633.  相似文献
